There are different types of boats to get to Nusa Penida, from public ferries to fast boats. Public ferries are generally cheaper but take more time, while fast boats are quicker but can be slightly more expensive. If you're prone to seasickness, it's advisable to take precautions before the trip, such as taking seasickness medication.
The boat schedules to Nusa Penida can vary depending on the ferry company and the season. Make sure to check the schedules in advance and plan accordingly. From my point of view, I suggest staying for at least a weekend because a single day is far too short considering the boat return.
I strongly recommend renting a scooter, as without it, you won't be able to do much. Scooter rental is very quick and hassle-free. Personally, I rented one shortly after arriving at the port; I paid, provided my phone number, and that was it. The next day, I returned the scooter to the port, and the owner came to pick it up, so it couldn't have been simpler!
However, be prepared for winding roads, ups, and downs as there are plenty !
As for accommodation, I recommend booking in advance. You have several options, ranging from rustic accommodations in wooden cabins to star-rated resort hotels.

Many ceremonies are held there, so if you're there during a ceremony, be mindful of your actions and respect the practitioners. You'll need to rent a sarong to cover your legs and shoulders before entering.
If your goal is to discover marine life and you prefer not to go alone to Nusa Penida, you also have the option of diving or snorkeling to observe the famous manta rays. In this case, I recommend booking your trip with Bali Breizh Divers. They speak English, French, and Indonesian, which will facilitate communication. You'll depart from Sanur in the morning by bus, then by boat to dive in this beautiful setting and return in the late afternoon.
